METHOD FOR DETERMINING THE CONTENT OF STABLE METABOLITES IN CELL CULTURE

Description of the development:
() Nitric oxide affects the cellular regulatory system, and therefore it is regarded as a signal molecule of many cellular processes. To quantify nitric oxide determine its stable metalobolity nitrite (NO2) and nitrate (NO3) in the dialysate tissues using the Griess reagent method Green. The novelty is that the definition of the content of stable nitric oxide is carried out in cell culture, which makes it possible to assess their immediate reaction, excluding the impact of the endocrine and nervous systems. To do this, mononuclear cells, such as healthy donors, pre-cultured for 24, 48 and 72 hours, reduce the volume and weight of the dialysate used reagents. In particular, up to 1 mL of effluent cell culture was added 0.1 ml of Griess reagent and 0.5 mL of effluent - 0.03 g of the reagent based on zinc dust, and 1 ml of dialysate was added 0.1 ml of Griess reagent, and before 0.5 mL of effluent 0.03 g of reagent-based zinc dust.
