METHOD FOR ACCELERATED QUALITY ASSESSMENT OF THAWED FROZEN BULL SEMEN
Purpose of the development:
Evaluation of quality indicators frozen-thawed semen sires.
Recommended application field:
Artificial insemination of cows.
Technical characteristic:
Improving quality assessment method involves incubating sperm in the semen sample and determination of biological ultrathermostat sperm motility, and used during the incubation temperature of 45 ° C, and the time intervals between the definition of mobility is 10 minutes.
Advantages over analogues:
Unlike the traditional method of determination of quality-thawed sperm (GOST 26030-83), by which the mobility rate is determined for 5 h, the proposed method allows almost 5 times reduce the scan samples of frozen sperm due to activation of metabolic processes within the cell. In general, certain indicators lasts no more than 1 h 40 min.
The development stage readiness:
Introduced in production
Technical and economic effect:
Method of rapid assessment of quality frozen-thawed semen makes it possible during the working day to verify the significant number (about 100) of sperm samples and can be applied in the breeding enterprises as a rapid test.
Description of the development:
() Semen doses (lined) were thawed at 38 ° C for further manipulation of the sperm sample using a thermostated syringe. In sperm gain preheated to 45 ° C the syringe. When you click a drop of semen is applied to a glass slide and transferred to a microscope with a heated table for the first evaluation. Return syringe with sperm in ultrathermostat at 45 ° C and every 10 minutes consistently performed quality assessment of sperm. Evaluation sperm is at 150 times magnification.
Tests carried out on the developed method based institute company purebred breeding in cattle and SE "Sumy breeding center."
