TRANSFER OF INNOVATIONAL TECHNOLOGIES

ORGANIC FUNCTIONALIZED SUPERPARAMAGNETIC NANOPARTICLES FOR REMOVAL OF PATHOLOGICAL CELLS


Purpose of the development: A new technology for the removal of pathological cells (apoptotic and necrotic) using lectin-bio-functionalized superparamagnetic nanoparticles.

Recommended application field: Removal of abnormal cells in the clinical diagnosis and treatment, scientific research.

Advantages over analogues: The high specificity of pathological cells in use of lectin as a "biosensor." High removal efficiency and separation of the cells. No damage to the cells. No need for expensive hardware.

Cell death can occur by two mechanisms - apoptosis or necrosis. Necrosis is a passive, pathological form of cell death, which is usually a response to external stress factors, such as inflammation, ischemic or toxic injury, and others. It does not happen under physiological conditions. In contrast, apoptosis is an active genetically programmed evolutionarily selected by cell death, and may occur as a pathological or under physiological conditions. Imbalance of apoptosis in the body can lead to serious illness from cancer (accumulation of neoplastic cells in the body) to immunodeficiency (loss of cell tissues and organs). Different cytomorphological and biochemical markers of apoptosis has been described in the cell. We have now found a novel biomarker development of apoptotic cell surface (plasma membrane). The choice of this biomarker for the detection of apoptosis does not require a violation of the integrity of the cell. The authors demonstrated that the plasma membrane of apoptotic cells contain high content of certain glycoproteins. Such changes have been proven to be a universal feature of apoptotic cells, regardless of their tissue origin or mode of inducing apoptosis. Specific lectins (carbohydrate binding proteins), it was proposed to use for the selective detection of proapoptotic glycoproteins. Various nanoparticles that were conjugated with a specific lectin used for removal of apoptotic cells from a mixed population. The new test lectin-induced agglutination was also developed for the rapid detection of apoptosis in cell samples examined in the clinic.
