METHOD OF THE BIOCOMPLEX REGISTRATION


Purpose of the development: Method for diagnosing human diseases and is based on fluorescent labeling of biomolecules colloidal quantum dots (QDs) based on A2B6 compounds. Attaching biomolecules to CT and, accordingly, this registration process is the first phase of such diagnosis.

Recommended application field: Medicine (diagnostics).

Advantages over analogues: Used for the registration process of education biocomplexes spectral CT radiation changes when acceding to biomolecules.

The proposed method is based on measuring the photoluminescence spectra of conjugated and unconjugated CT and uses the effect of spectral changes conjugated QD luminescence compared to the unconjugated. Currently biocomplexes registration using CT scans performed on the photoluminescence intensity. However, in this case the accuracy of the analysis can be low because the reaction usually proceeds biokonyugatsii not fully in solution and can remain unconjugated RT. The effect of changes in the spectral photoluminescence of conjugated CT allows us to offer a new method for detecting bio-complexes, which can improve the accuracy of the analysis. To implement changes to effect droplet spectral conjugated and unconjugated CT were deposited on solid solution type crystalline Si substrate or SiC and dried at 50 ° C. In this case the spectral position of the maximum bandwidth of conjugated CT shifted to shorter wavelengths in comparison with the position of the strip of unconjugated CT, which was manifested in a clearly visible change in emission color. The offset value can be up to 50 nm. The method was tested on a commercial CT CdSe96Te04 ZnS shell polymer coated with a radiation wave length 705 nm. Part was combined with biomolecules CT - commercial monoclonal antibodies: IL-10, IL-6, OPG, CAV-1 and P53, PSA, and conjugation procedure was carried out using a commercial kit for RT conjugation with a wavelength of 705 nm.
